Built around a gold nanoparticles, Au079 is designed to allow cancer therapies to more efficiently target the tumor.

By treating solid tumors with immune modulating cytokines in previously impossible-to-dose drug combinations, we are able to activate an immune response and disrupt the vasculature of the tumor.

Clinical highlights

Exceeded safety thresholds: The formulation has demonstrated the ability to surpass the Maximally Tolerated Dose (MTD) of TNF without dose-limiting toxicities typically associated with TNF, such as hypotension and hepatotoxicity.

Enhanced TNF exposure: Increases systemic exposure to TNF by up to 40- fold compared to conventional formulations.

Innovating on the Au079 Platform

Building on the Au079 platform: Advancements in clinical formulation are further refining the balance between potency and safety.

Increased TNF payload: Novel formulations of Au079 have been developed to boost TNF particle content by 2-3 times without compromising the stealth properties of the nanoparticles.

Optimized PEG-THIOL composition: Modifications in the composition of PEG-THIOL have unlocked new possibilities:

  • Variable PEG-THIOL chain lengths: Using a combination of “shorter” or “taller” PEG-THIOL molecules allows for more efficient TNF binding and accessibility.
  • Introduction of PolyPEG: This approach offers the potential to unmask TNF molecules otherwise shielded by the 20 kDa PEG-THIOL, enhancing therapeutic engagement with tumor targets.

By leveraging these innovations, the platform continues to push the boundaries of nanotechnology-based cancer therapy, enhancing the effectiveness of TNF while maintaining a favorable safety profile.
